主要研究方向

  

1.    四链核酸结构与功能研究及分析应用:设计开发折叠可控的双分子G-四链体与i-motif结构,用于DNA纳米组装及可开关传感分析;研究四链核酸与荧光配体作用情况,发展新型信号读出体系,用于免标记生物分析及细胞成像;研究四链核酸引导金纳米、金棒组装与等离子体增强情况,拓展其活体应用前景。

2.    DNA纳米机器设计与应用:以DNA四面体等框架核酸为纳米脚手架,利用G-四链体、i-motifATP适配体等响应外界刺激,发展细胞微环境识别的智能型DNA逻辑电路;将DNA电路与纳米机器进行有机整合,在细胞、动物活体内原位控制分子载荷靶向传递与精准释放,进行microRNAmRNA成像,抑制疾病基因表达。

3.    球形核酸酶(SNAzyme)催化的电化学发光及疾病检测应用:基于G-四链体与血红素在金纳米核表面组装,制备高活性、高稳定性的球形核酸酶,结合核酸放大技术,将球形核酸酶催化的电化学发光体系用于疾病相关核酸标志物灵敏检测;利用框架核酸搭建电化学发光纳米平台,对循环microRNA、外泌体RNA进行增强检测。
